Discovery of C-9 Modified Berberine Derivatives as Novel Lipid-Lowering Agents

نویسندگان

چکیده

Berberine (BBR), a kind of quaternary ammonium benzylisoquinoline alkaloids with multiple pharmacological activities, has been regarded as promising lipid-lowering agent in the field drug repurposing. Particularly, chemical modification at C-9 position BBR can remarkably improve its efficacy. In this study, thirteen novel derivatives were rationally designed, synthesized, and evaluated by preliminary tests. The results showed that most compounds exhibited more potent hypolipidemic activities when compared simvastatin. Among these compounds, compound 2h-1 2h-2 better activity profiling four tests involving inhibition total cholesterol (TCHO), triglyceride (TG), low-density lipoprotein (LDLC) increase high-density (HDLC). Correspondingly, analogs 9-O-cinnamic moiety probably activity, should be exploited an important versatile template for development BBR-like agents.

Novel 5-substituted-1H-tetrazole derivatives as potent glucose and lipid lowering agents.

A series of 5-(4-alkoxyphenylalkyl)-1H-tetrazole derivatives, containing an oxazole-based group at the alkoxy moiety, was prepared and their antidiabetic effects were evaluated in two genetically obese and diabetic animal models, KKAy mice and Wistar fatty rats. Syntheses were performed by cyclization of the corresponding nitrites reacting with azide compounds. A large number of the 5-(4-alkoxy...

Novel Levofloxacin Derivatives as Potent Antibacterial Agents

In this report, a new series of fluoroquinolone agents which was derived from levofloxacin was synthesized and evaluated against Gram-positive (Bacillus subtillis, Staphylococcus aureus, Staphylococcus epidermidis and Micrococcus luteus) and Gram-negative (Esherichio coli, Klebsiella pneumoniae, Pseudomonas aeruginosa and Serratia marcescens) organisms. The results showed that some of the synth...

Novel Uses for Lipid-Lowering Agents

Statin use leads to a reduction in the downstream products of the mevalonate pathway. Knowledge of this pathway has led scientists to investigate the role of statins in cancer prevention and treatment. Statins appear to possess a variety of pleiotropic effects, including inhibition of cell proliferation; enhanced apoptosis; and modulation of inflammation, endothelial function, and angiogenesis....

Discovery of novel taspine derivatives as antiangiogenic agents.

VEGFR-2 plays a critical role in vasculogenesis and inhibitors of VEGFR-2 could be used in the treatment of cancer. Taspine was one of the active ingredients screened by using an endothelial cell membrane chromatography and showed inhibition against VEGFR-2. In our research, we explored how the lactone ring and biphenyl scaffold in taspine influence its potent in vitro anticancer and antiangiog...

We previously reported that (Z)-2-(4-[(5-methyl-2-phenyl-1, 3-oxazol-4-yl)methoxy]benzyloxyimino)-2-(4-phenoxyphenyl)acetic acid (3) showed potent glucose and lipid lowering effects in genetically obese and diabetic mice, KKA(y). This compound also showed transcriptional activity for peroxisome proliferator-activated receptor (PPAR)-gamma. We expanded on the structure-activity relationships of ...
